Histopathological Study on he Histogenesis of Pulmonary Adenoma Induced by Urethane in Mice

Abstract
To study the histogenesis of pulomonary adenoma which is quite similar to bronchioloaoveolar carcinoma od human lung. Histopathological examination was done after repeated administration of urethane to the 3rd week old weaning mice and single administration to prenatal mice. Animals were sacrificed sequentially at 2nd, 3rd, 4th, and 7th month after exposure to urethane. The results octained were as follows: 1. Multifocal and active proliferation of alveolar cells were seen in the peripheral and central portion of the lung since 2 months after first administration of urethane without proliferation of bronchiolar cells. Peripheral portion was more affected than central portion. More numerous lesions of active proliferation and adenomas were noted in repeated multiple administration group than single exposed preantal group. 2. Small adenomas about 1 mm in diameter were formed since 3rd month after first administration of urethane and progressively enlarged to 6 mm in diameter 7th month. Peripheral portion was more affected than central portion of the lung. 3. Focal proliferation of nonciliated bronchiolar cells(Clara cells) was seen in only one mouse with out evidence of progression to tumefaction. 4. Therefore, the pummonary adenoma induced by urethane was developed multifocallt through active proliferation of alveolar cells along the alveolar septae accompanying atypical nuclear to form pulomonary adenomas with peropheral prediliection. So the type cell of pulmonary adenoma was to be alveolar cell of the lung.
